Electrophoretic Titration Curve (ET)

  • Electrophoretic titration (ET) curves for crude venoms and fractions are performed to determine optimal conditions for separation by ion exchange chromatography, and to evaluate the pI of venom proteins.

  • A volume of 3.5 µl (5.25-20 µg) sample is separated by electrophoretic titration using a Pharmacia Biotech PhastSystem.

  • A pH gradient of 3-9 is established on a polyacrylamide isoelectric focusing (IEF 3-9) PhastGel.

  • The gel is rotated 90˚ and the venom sample applied. The gel is stained with silver nitrate.
